Can you defrag a computer on a network?

I have 3 dell computers networked together and I was wondering if you can defrag any of the computers while connected to the network, or just the main server, or do I shut down any of the computer while defrag?

In a word, "NO". You need to disconnect the machine you intend to defrag, there shouldn't need to be shut down the network though, simply disconnect the network cable and suspend the task manager.
