Retaining nut under kitchen tap cover?

Should the retaining nut under the cover of a kitchen tap be turned in the direction as any ordinary screw thread. I am working on a very stiff nut and am being careful not to try too hard in the wrong direction.

Yes they are the normal right hand threads. They are often stiff due to limescale so you'll need a bigger spanner, but do take care that the tap itself doesn't move at all or you'll risk a water leak underneath.
